Eksplorasi Isolat Bakteri Rizosfer Musa acuminata Pada Fermentasi Ampas Tebu Sebagai Kandidat Biofertilizer

Rizosfer tanaman adalah lingkungan mikroorganisme yang memiliki kemampuan besar sebagai sumber bakteri fungsional untuk mendukung praktik pertanian yang berkelanjutan. Tujuan dari penelitian ini adalah untuk mengkarakterisasi isolat bakteri dari rizosfer Musa acuminata, menilai kemampuan mereka dalam fermentasi ampas tebu, dan mengeksplorasi potensinya sebagai calon biofertilizer berdasarkan produksi Asam Indole Asetat (IAA). Penelitian ini dilakukan melalui eksperimen di laboratorium dengan metode isolasi bakteri menggunakan media Nutrient Agar (NA), pemurnian isolat dengan metode streak plate, pewarnaan Gram, uji produksi IAA menggunakan reagen Salkowski, serta fermentasi ampas tebu selama 14 hari dan menghitung viabilitas bakteri menggunakan metode Total Plate Count (TPC). Hasil penelitian mengungkapkan bahwa isolat bakteri memiliki morfologi koloni yang berwarna putih krem, berbentuk bulat, dengan elevasi cembung dan tepian yang rata. Pewarnaan Gram menunjukkan bahwa bakteri tersebut bersifat Gram positif dan berbentuk sel kokus yang berkelompok. Uji IAA menghasilkan perubahan warna menjadi merah muda setelah penambahan reagen Salkowski, yang menandakan adanya produksi IAA secara kualitatif. Fermentasi ampas tebu membawa perubahan karakteristik fisik, termasuk peningkatan kekeruhan, munculnya aroma asam, serta terbentuknya endapan setelah inkubasi selama 14 hari. Perhitungan TPC pada pengenceran 10⁻⁶ menunjukkan ada 34 koloni dengan total viabilitas sebesar 3,4 × 10⁸ CFU/mL. Dengan demikian, isolat bakteri dari rizosfer Musa acuminata menunjukkan potensi awal sebagai kandidat biofertilizer yang berbasis fermentasi ampas tebu.
